What are the recommendations for samples with different cell diameters?

Answer:
To ensure accurate counting results, samples should be allowed sufficient time for cells or nuclei to settle to the same focal plane after the counting chip is inserted into the instrument.

Recommended settling times are as follows:

  • Samples with average diameter ≥10 μm (e.g., cultured cells):
    Allow 30 seconds to 1 minute before measurement 
  • Samples with average diameter between 5–10 μm (e.g., nuclei):
    Allow 1.5 to 2 minutes before measurement 
  • Samples with diameter <5 μm:
    Longer settling times may be required to ensure proper focusing and accurate detection 

These recommendations help ensure that all particles are positioned within the imaging plane, improving counting accuracy.
