The SeekOne DD 3' scRNA-seq workflow is compatible with a variety of sample types, including:
- Fresh tissues
- Frozen tissues (via nuclei isolation)
- Cell suspensions
- Nuclei suspensions
- Plant protoplasts
The recommended particle size range is 5–40 μm.
Detailed guidance is provided below:
- Fresh tissues:
Strongly recommended. Samples should be processed as soon as possible after collection to preserve cell viability. - Frozen tissues:
Must be snap-frozen in liquid nitrogen. Only nuclei can be used for downstream processing.
- Cell suspensions:
Recommended quality metrics include:- Viability >90%
- Aggregation rate <10%
- Nucleated cell proportion >70%
- Viability >90%
- Nuclei suspensions:
Suitable for large cells or tissues that are difficult to dissociate (see Q3). - Plant protoplasts:
Applicable after removal of the cell wall.
Important:
This workflow is not compatible with FFPE samples.
