What's Hiding in the Half of the Transcriptome That Single-Cell 3′ Sequencing Misses

Full-Length Transcriptome | Mainstream single-cell 3′ sequencing only reads the last stretch of a transcript — enough to count genes, but not enough to see what's written in between. Alternative splicing, fusion breakpoints, mutation sites, and every RNA born without a poly(A) tail all fall outside its reach. This piece looks at what a single primer swap — from oligo-dT to a semi-random primer — brings back into view, and why the missing half of the transcriptome may hold more answers than the half we've been reading all along.

Beyond Elimination: How Single-cell Analysis Revealed a New Way to Control Senescent Cell Behavior

Using SeekOne™️ DD single-cell 3’ RNA sequencing, researchers found that abemaciclib (a CDK4/6 inhibitor already approved for breast cancer) doesn't kill senescent cells or reverse their arrested state. It selectively quiets NF-κB-driven inflammation, while leaving other senescence programs untouched. Even more unexpected: CDK4/6 turn out to have a second job. In cells that have permanently stopped dividing, they still operate — this time as part of a regulatory complex with NF-κB and the retinoic acid receptor RARα. Break that complex, and the inflammatory signal drops with it.
