Yes.
The SeekOne DD Multiome Methylation + RNA workflow supports downstream enrichment of full-length immune receptor V(D)J sequences from the same single cells, including TCR (T-cell receptor) and BCR (B-cell receptor).
Following cDNA secondary amplification, the purified cDNA product generated in Step 4–5 can be directly used for V(D)J enrichment workflows.
The downstream enrichment procedure follows the same workflow described in the SeekOne DD Single-Cell 5′ Transcriptome and Immune Profiling Kit protocol.
The dedicated V(D)J enrichment kits must be purchased separately.

Important notes
- V(D)J enrichment is performed using amplified cDNA generated from the same cells used for methylation and transcriptome profiling.
- Separate immune repertoire library preparation is required.
- RNA quality and cell viability remain important factors affecting V(D)J enrichment performance.
V(D)J enrichment requires whole-cell input and is not supported for isolated nuclei samples. This is because mature TCR and BCR transcripts are predominantly localized in the cytoplasm and may not be sufficiently preserved in nucleus-only preparations.
