What sample types are compatible with scMLPSeq?

Answer:
scMLPSeq currently supports the following sample types:

1. Human FFPE samples

FFPE tissues must first be processed into nuclei suspensions according to the SeekMate FFPE Dissociation Kit V2.0 User Manual before proceeding with the scMLPSeq workflow.

2. 1% PFA-fixed human cells or nuclei suspensions

The workflow supports:

• Cell suspensions fixed with 1% PFA at room temperature for 10 min 
• Nuclei suspensions fixed with 1% PFA at room temperature for 10 min 

Applicable input materials include:

• Cells or nuclei isolated from fresh samples 
• Nuclei isolated from frozen samples 

scMLPSeq is a probe hybridization-based workflow compatible with fixed samples. Appropriate fixation helps preserve RNA localization and cellular structure which may reduce RNA loss during sample processing. However, fixation conditions must be carefully controlled. Excessive fixation may reduce probe accessibility, hybridization efficiency and ligation-extension efficiency. 

Therefore, only the conditions described above are currently validated.
