What should I do if cDNA yield is low after V(D)J enrichment?

Answer:
PCR cycles may be increased, but the total number of cycles should not exceed 14.

Pre-checks:

  • Check T/B cell proportion (recommended >20%)
    Low abundance directly reduces enrichment efficiency 
  • Check sample quality: Fresh samples perform better
    Frozen samples often show lower V(D)J yield due to RNA loss or degradation 

If still low after optimization: Recommend re-preparing with higher-quality samples
