How can RNA degradation be minimized?

Answer:
To preserve native transcriptional states as effectively as possible:
- Remove visibly necrotic or calcified regions whenever possible.
- Freeze tissue immediately after collection.
- Avoid prolonged storage on ice.
- Avoid repeated freeze-thaw cycles.
- Store samples at −80°C for long-term preservation.
For tissues that cannot be embedded immediately:
- Place the tissue into cryovials first.
- Store at −80°C.
- Perform OCT embedding later under low-temperature conditions such as dry ice.
Important notes
- Long-term storage at −20°C is not recommended.
- Frequent retrieval from liquid nitrogen storage may also introduce temperature fluctuations that negatively affect RNA stability.
