Within a single capture area (5.5 mm × 15 mm), the tissue cross-sectional area should range from:

• Minimum: 3 mm × 3 mm
• Maximum: 5.5 mm × 15 mm

The region of interest should have a thickness greater than 200 μm.

Based on previous experience, most routine tissue samples are typically controlled within approximately 5 mm × 5 mm.

The elongated capture area design allows multiple tissue samples to be processed on the same chip, helping reduce per-sample experimental cost.

Recommendations for multi-section layouts

If performing multi-section layouts:

• Tissue sections should ideally share similar pathological structures.
• H&E staining characteristics should be as consistent as possible.
• Regions of interest should remain on the same sectioning plane.

These recommendations are important because different tissue types may exhibit differences in RNA release efficiency, nuclei isolation efficiency and spatial barcode capture efficiency. Such variability can reduce data uniformity across regions.
